Basic Info

What is N-Acetyl-L-aspartic acid?

N-acetylaspartic acid or N-acetylaspartic acid (NAA) is a by-product of aspartic acid, with molecular formula of C6H9NO5 as well as molecular weight of 175.139.

NAA is the 2nd highest possible focus particle in the brain, second just to amino acid glutamate. It was discovered in adult minds of nerve cells, oligodendrocytes as well as myelin, and was manufactured by the amino acid aspartic acid and acetyl coenzyme A in mitochondria.

N-acetyl-L-aspartic acid, abbreviated as L-N-AcAsp, is an amino acid derivative, in which the acyl group is designated as the acetyl group. Acetylation of L-aspartic acid is achieved by first neutralizing with potassium carbonate to pH 7.0, and then adding acetic anhydride. After the hydrochloric acid acidification product is used, the acetylation product obtained is purified by HPLC.

N-Acetyl-L-aspartic acid Uses

NAA may act as a neurotransmitter in the brain by acting on metabolic glutamate receptors.

NAA may be a sign of creativity. The high level of NAA in the hippocampus is related to the better performance of human working memory.

N-acetyl-L-aspartate is used in the biological research of metabolic changes related to schizophrenia.

N-acetyl-L-aspartic acid can be synthesized as a reactant:

  • Protection of homoserine by selective reduction and acid-catalyzed cyclization γ- Lactone.
  • The racemic amino-substituted succinimide derivatives were obtained by ring condensation reaction.

Application of N-Acetyl-L-aspartic acid

Various functions provided by NAA are under investigation, but the main proposed functions include:

  • Neuron penetrant involved in brain fluid balance.
  • The acetate source of lipid and myelin synthesis in oligodendrocytes, which are neuroglial cells myelinated by neuronal axons.
  • Precursor of synthetic neuropeptide N-acetylaspartate glutamate.
  • Contribute to the energy production of amino acid glutamate in neuron mitochondria.

In the brain, NAA is believed to exist mainly in the neuronal cell body, where it acts as a neuronal marker, but it can also spread freely to the entire nerve fiber.

CN201710830763.2 reports a method and its application of using diagnostic markers to evaluate the early toxicity of trace exogenous chemicals.

A method for evaluating the early toxicity of trace exogenous chemicals using diagnostic markers. The diagnostic markers include: L-valine, N-acetyl-L-aspartic acid, lysolecithin, creatinine, and 2-phosphoglycerol Any one or more of acids. In the process of studying exogenous chemicals, it was found that exogenous chemicals can disrupt the amino acid balance in zebrafish and affect the catabolism of proteins. This disturbance in the developmental stage is associated with its toxicity mechanism. The inventors found that the zebrafish The combination of modeling and metabolomics techniques can greatly improve the sensitivity of toxicity prediction methods.

Therefore, the changes of the above-mentioned zebrafish metabolite indicators can be used to judge the early toxicity of exogenous chemicals.
