Baclofen, sold under the brand name Lioresal, among others is a central nervous system depressant used as a skeletal muscle relaxant. It is primarily used to treat spasticity. It is also used as a muscle relaxant by compounding pharmacies in topical pain creams.
It is a GABA receptor agonist, specifically of the GABAB receptors. Its beneficial effects in spasticity result from actions at spinal and supraspinal sites. It is a derivative of γ-aminobutyric acid (GABA).
A beneficial property of baclofen is that tolerance to its muscle-related therapeutic benefits does not seem to occur to a significant degree – baclofen retains its therapeutic anti-spasmodic effects even after many years of continued use. Newer studies, however, indicate that tolerance may develop in some people receiving intrathecal baclofen treatment. As of 2015 the cost for a typical course of treatment in the United States is less than 25 USD.



Production methods
The ethyl methyl – diacetyl acetate was formed by condensation of chlorobenzaldehyde and ethyl acetoacetate. The hydrolysis of heated water is applied to the chlorphenyl glutaric acid. Chlorinated phenyl glutaric anhydride was formed by anhydride dehydration. In the end, the chlorophenyl-butyric acid was decomposed by using a strong ammonia amine. The final open loop and degradation are performed as follows: 42.25g beta – (for chlorphenyl) glutaramide, which is stirred and added to the 200ml aqueous solution containing 8.32g of sodium hydroxide. Mixture in 10 min 50 ℃ heating, cooling and 10-15 ℃, add 200 ml water solution containing 40.9 g of sodium hydroxide, after 20 min, add 38.8 g bromine, stir in 20 to 25 ℃ 8 h, will carefully with concentrated hydrochloric acid reaction liquid pH value to 7, then the settling of fine beta – (on) – gamma amino butyric acid crystallization, again by recrystallization in water, melting point 206-208 ℃.
